Flowering time data for X5 and X6 Biomap trials is complete and uploaded to /Drive/2015_Field_Files/phenotype_files/

16 DAP destructive sampling collections were done from 8/10/15 through 8/20/15. Details of samples collected on each date can be found in /2015_Field_Files/phenotype_files/distructive_sampling_pollination_notes_080415.xlsx

For each genotype, three replicates were harvested that each consisted of tissue from three plants. Endosperm and embryo tissue from 50 kernels per ear (150 total for each replicate) were pooled to make up a single replicate. When only two ears were available, 75 kernels from each ear were dissected and pooled to make 150 total for the replicate. Internode tissue from the internode above the ear bearing node was also harvested from the same three plants.

Flowering time data for the IGXE plot in X6 is complete and uploaded to /2015_Field_Files/phenotype_files/X6_IGXE_floweringtime.xlsx

The fourth round of R1 field destructive sampling was done from 2:00-2:30 on 08/03/15.

The final three genotypes were ready today (LH185, MoG, PHJR5). For each genotype 3 replicates were harvested with the exception of PHJR5 which only had 1 replicate that needed to be harvested. Each replicate contained three plants at the R1 developmental stage. The tassel (anthers) were harvested. LH185 was harvested from a block in X5 (SP15:10146-10155), MoG was harvested from the block in X6 (SP15:20673-20720), and PHJR5 was harvested from the yield component experiment in X5.

These samples are all in labeled bags in the -80 freezer.

The third round of R1 field destructive sampling was done from 2:30-3:00 on 07/30/15.

Six of the genotypes were ready today (W22, B73, PHK76, PHK56, PHK35, PHJR5). For each genotype 3 replicates were harvested with the exception of PHJR5 which only had 2 replicates that were ready. Each replicate contained three plants at the R1 developmental stage. The tassel (anthers) were harvested. W22 was harvested from the block in X6 (SP15:20673-20720), B73 was harvested from the extra females range in the back range of X6, PHK76 and PHK56 were harvested from the second (delay) block in X7 (SP15:30169-30192), PHK35 was harvested from the yield component experiment in X6, PHJR5 was harvested from the yield component experiment in X5.

These samples are all in labeled bags in the -80 freezer.

NOTE: I had a note that we collected PHK56 in the second harvest, but we believe this was really PHP02 and that the notes that I took were incorrect.

The fourth round of V15 field destructive sampling was done from 1:30-3:00 on 07/28/15.

The final genotype was ready today (MoG). Three replicates were harvested. Each replicated contained three plants at the ~V15 developmental stage (Note: the bags say V18 as we thought this would be the stage when the plants would be ready, but on average they are at V15). The immature tassel and immature ear were harvested. MoG was collected from the block in X6 (SP15:20703).

These samples are all in labeled bags in the -80 freezer.

Three of the genotypes were ready today (PHK56, PHN46, and W606S). For each genotype 3 replicates were harvested. Each replicate contained three plants at the R1 developmental stage. The tassel (anthers) were harvested. PHK56 was harvested from the first block in X7 (SP15:30145-SP15:30167) and PHN46 and W606S were harvested from the block in X6 (SP15:20673-20720).

These samples are all in labeled bags in the -80 freezer.

The third round of V15 field destructive sampling was done from 1:30-3:00 on 07/23/15.

Four of the genotypes were ready today (LH185, PHK76, PHK35, and PHJR5). For each genotype 3 replicates were harvested. Each replicated contained three plants at the ~V15 developmental stage (Note: the bags say V18 as we thought this would be the stage when the plants would be ready, but on average they are at V15). The immature tassel and immature ear were harvested. LH185 and PHK76 were collected from the second (delay) block in X7 (SP15:30169-30192), PHK35 was harvested from the yield component trial in X6 from rep 1 (YC15:1324) and PHJR5 was harvested from the yield component trial in X5 from rep 4 (YC15:4240).

Four of the genotypes were ready today (LH82, DK3IIH6, PH207, and PHJ89). For each genotype 3 replicates were harvested. Each replicate contained three plants at the R1 developmental stage. The tassel (anthers) were harvested. Everything was harvested from the first block in X7 (SP15:30145-SP15:30167).

These samples are all in labeled bags in the -80 freezer.

The second round of V15 field destructive sampling was done from 1:30-3:00 on 07/20/15.

Six of the genotypes were ready today (DK3IIH6, LH82, PHK56, PHN46, W606, B73). For each genotype 3 replicates were harvested. Each replicated contained three plants at the ~V15 developmental stage (Note: the bags say V18 as we thought this would be the stage when the plants would be ready, but on average they are at V15). The immature tassel and immature ear were harvested. All samples were collected from the first block in X7 (SP15:30145-30167) except for B73 that was harvested from SP15:20612.

Four of the genotypes were ready today (PH207, PHJ89, PHP02, and W22 (a little bit early, but still sampled)). For each genotype 3 replicates were harvested. Each replicated contained three plants at the V15 developmental stage (Note: the bags say V18 as we thought this would be the stage when the plants would be ready, but on average they are at V15). The immature tassel and immature ear were harvested. All samples were collected from the first block in X7 (SP15:30145-30167).

Collars were put on between the 10th and 11th leaves in the hybrid plots on 07/08/15 and the inbred plots on 07/14/15 of the second St Paul planting of the biomap experiment.

HGXE trail had no issues aside from a clogged rain catcher on the weather station. Plants were V9-V10. The station was cleared of debris and confirmed as working.

Planting 1 had some known issues that occurred during planting. Plots below had no plants growing in them, and no sign of ever having any emerged plants:

The V11 field destructive sampling was done from 9:00-11:30 on 07/02/15.

For each genotype (DK3IIH6, LH185, LH82, MoG, PH207, PHJ89, PHK56, PHK76, PHN46, PHP02, W22, W606S, B73, PHJR5, PHK35) 3 replicates were harvested. Each replicated contained three plants at the V11 developmental stage. The first leaf without a ligule initiated was dissected for each plant (~13 leaf) and the base (papery white tissue), light green tissue in the middle, and dark green tissue at the tip of the leaf was harvested.

All samples were collected from the first block in X7 (SP15:30145-30167) with the exception of LH185 reps 2 and 3 were harvested from X6 back (SP15:20120-20182), B73 reps 1-3 from X6 back (SP15:20610), PHJR5 reps 1-3 from X6 yield component trial (YC15:2038), and PHK35 reps 1-3 from X6 yield component trial (YC15:2329).

Rep1 of the BiomAP inbred/hybrid greenhouse experiment was measured and harvested on 6/26/15 from 8:00-2:30. Samples were placed in the oven at 85F from 6/26/15-6/26/15.

Rep2 of the BiomAP inbred/hybrid greenhouse experiment was measured and harvested on 6/29/15 from 8:00-1:00.

Placed Biomap Greenhouse planting samples in Drier #1 (140°) shelf #2 at 2:00pm on June 29, 2015.

5th leaves were punched for the BiomAP inbred/hybrid trial in X5 on 06/22/15.

The 6 DAS greenhouse destructive sampling was done from 9:30-11:30 on 6/22/15.

For each genotype (DK3IIH6, LH185, LH82, MoG, PH207, PHJ89, PHK56, PHK76, PHN46, PHP02, W22, W606S, B73, PHJR5, PHK35) 3 replicates were harvested. Each replicated contained three plants at the 6 DAS developmental stage with the exception of LH185 that only had two plants per replicate.

There were two plants (one from PHN46 rep2 and one from PHK76 rep2) that looked stressed and were not harvested. These plants had white, yellow, and pink coloring in the leaves.

These samples are al in labeled bags in the fourth shelf of the -80 freezer.

Rep2 of the BiomAP inbred/hybrid greenhouse experiment for measuring seedling root length, above ground height, root weight, and above ground weight was planted on 6/22/15. The greenhouse conditions are 16h day/8 h night with supplemental light only as needed and temperatures are at 27C day/24C night.

X5, X6, and X7 were all sprayed with atrazine on 6/8/15 to help with emerging grass issue. The fields were rechecked on 6/17/15 and look good.

The inbreds for destructive sampling in the greenhouse at 6 DAS were soaked overnight on the night of 6/15/15 and planted on 6/16/15. The greenhouse conditions are 16h day/8 h night with supplemental light only as needed and temperatures are at 27C day/24C night.

The final experimental file was updated on 6/16/15 to change the boarder pots from B73 to Mo17.

Stand counts on all X6 plots except HOF/NIL were collected on 6/4/15 and are available here.

The first phenotyping by blueriver was done on 6/8/15 and including the entire front section of X6.
